The goal of this study is to explore the effects of three known regulation proteins of Paracoccus denitrificans proteome during adaptation to changed environmental conditions. It is known that these proteins serve as sensors for oxygen and nitrogenous terminal electron acceptors. After construction of the sequence database for mass spectrometry protein identification based on published P. denitrificans genome sequence, a detailed complex proteomic analysis will be performed. The effects of mutation in genes for regulation proteins on the expression of other proteins in total cell lysates will be examined. The proteins will be separated by two-dimensional gel electroporesis and identified by mass spectrometry. The proteins with changed expression will be analyzed by bioinformatic tools, the functional analyses will be performed for selected proteins. A new chromatographic/mass spectrometric method will be tested for a comparison of membrane fractions protein composition. A created sequence database will be also applied for identification of proteins from previously published studies. The results obtained will be used for supplementation of public 2-D P. denitrificans proteome database (http://www.mpiib-berlin.mpg.de/2D-PAGE/).
