Procathepsin D (pCD) is over-expressed and secreted by several types of tumors and cancer cell lines. Increased level of pCD is known to affect multiple stages of tumor progression, including proliferation, invasion and metastasis and is associated with poor prognosis of various cancers. It has been suggested that protein-protein interactions are important mediators of these effects. Using gene microarray approach we have recently identified several differentially expressed genes upon treatment of breast cancer cells with pCD. This project is therefore designed to A) explore the role of NF-kB2 and MAPK in mediating the effects of pCD on breast cancer cells, B) search for pCD protein-protein interacting partners, C) analyze the role of pCD-mediated signaling in regulation of apoptosis of breast cancer cells. We believe that this project can answer the question how the pCD affects cancer cells. Full understanding of this mechanism is important for design of pCD-targeted cancer therapy.
